Assessment of fluorescein diacetate hydrolysis as a measure of total esterase activity in natural stream sediment biofilms

چکیده

I applied the fluorescein diacetate (FDA) hydrolysis technique as a rapid and sensitive estimator of total esterase activity in stream sediment biofilms. I investigated the effects of temperature, pH and incubation time and optimized the assay for low blanks and high fluorescein extraction. The FDA procedure was precise (c.v. = 4.15%) and could detect 10 nM fluorescein. Spatial patterns of esterase activity within stream sediment biofilms correlated with electron transport system activity, bacterial thymidine incorporation, glucosidase activity and chlorophyll a. As such, I suggest he modified and optimized technique as applicable to the investigation of total stream bioflim esterase activity.

Fluorescein diacetate hydrolysis as a measure of total microbial activity in soil and litter.

Spectrophotometric determination of the hydrolysis of fluorescein diacetate (FDA) was shown to be a simple, sensitive, and rapid method for determining microbial activity in soil and litter. FDA hydrolysis was studied in soil and straw incubated for up to 3 h. Hydrolysis was found to increase linearly with soil addition. FDA hydrolysis by pure cultures of Fusarium culmorum increased linearly wi...

Development of a whole-sediment toxicity test using a benthic marine microalga.

An acute whole-sediment toxicity test with a benthic marine microalga was developed and optimized using flow cytometry to distinguish algae (based on their chlorophyll a autofluorescence) from sediment particles. Of seven benthic marine algae screened, the diatom Entomoneis cf punctulata was most suitable because of its tolerance of a wide range of water and sediment physicochemical parameters,...

Assay for fluorescein diacetate hydrolytic activity: Optimization for soil samples

With the increased interest in integrated soil bioecosystem studies, there is a need to have a method of measuring overall microbial activity potential. Hydrolysis of fluorescein diacetate [3 0,6 0-diacetylfluorescein (FDA)] has been suggested as a possible method because the ubiquitous lipase, protease, and esterase enzymes are involved in the hydrolysis of FDA. Following hydrolysis of FDA, fl...

Development of a sensitive and rapid method for the measurement of total microbial activity using Fluorescein Diacetate (FDA) in a range of soils

Fluorescein diacetate (FDA) hydrolysis is widely accepted as an accurate and simple method for measuring total microbial activity in a range of environmental samples, including soils. Colourless fluorescein diacetate is hydrolysed by both free and membrane bound enzymes, releasing a coloured end product fluorescein which can be measured by spectrophotometry. The current method for measuring FDA...

Age dependency of esterase activity in rat and human keratinocytes.

The age and species dependent characteristics of cutaneous esterase activity were examined in cultured keratinocytes of neonatal and adult humans and of rats at the age of 1, 3, 10, and 50 d. The existence of esterases was characterized using fluorescein-5-isothiocyanate diacetate under a confocal laser scanning microscope. In vitro hydrolysis of ethyl nicotinate (EN), an esterified prodrug of ...
